19. Cultures can hereafter be routinely maintained by repeating steps 16 & 17. Equipment a. Healthy, greenhouse-grown tobacco plants ca. 5-1 m tall. b. Sharp scalpel. c. Sterile scalpel, long- and short handled forceps which, when not in use are stored with the ends in 70% EtOH. Before reuse the alcohol is removed by flaming. d. Non-sterile 9 em Petri dishes containing moistened filter paper. e. Molten wax. f. 250 ml beaker containing ca. 150 ml 70% (v/v) EtOH in water. g. 2 sterile 250 ml beakers.

Equipment: a. Established, friable callus cultures. b. Autoclaved, 250 mi. wide-necked Erlenmeyer flasks containing 50 ml culture medium and sealed with a double layer of aluminium foil. c. A supply of sterile aluminium foil sheets 10 X 10 em. PTCM-A3/9 d. Sterile spoon spatula which, when not in use, is stored with the end in 70% EtOH. Before using, the alcohol is quickly removed by flaming. e. tm nylon or stainless steel filters + glass filter funnels. f. Sterile 100 ml Erlenmeyer flasks. g. Sterile wide-mouthed glass pipettes.

It is advisable, especially for inexperienced workers, to have an extra supply of sterile aluminium foil sheets available to use when the originals are accidentally torn on removal from the flasks. 3. If the callus is not characteristically "crumbly" the outer, more friable regions can be gently scraped/sliced off for use as the inoculum. No callus showing signs of browning should be transferred. 5. Use speeds of 90-110 rpm with a throw amplitude of ca. 1 em. Incubation at 25 ° C is preferred.
